| colorectal cancer | We investigated the wide variability of 18F-2-fluoro-2-deoxy-D: -glucose (FDG) uptake, semiquantified as standardized uptake value (SUV), in positron emission tomography (PET) scanning, in 20 patients with colorectal cancer (CRC), including 1 with synchronous hepatic metastasis. The sensitivity of PET in CRC diagnosis was 100%, with a mean SUV of 8.0 (3.1-11.9). tumor size and depth of invasion were associated with higher SUVs (P=.0004, .042, respectively). Strong glucose transporter-1 (GLUT-1) expression had significantly positive correlation with the SUV (r=.619, P=.003). GLUT-1 expression revealed positive staining in 17 (85%) of the 20 primary lesions. The central part of the tumor, thought to be relatively hypoxic, had stronger GLUT-1 expression and a higher SUV than the periphery, in both the primary tumor and hepatic metastatic foci. Our data suggest that the SUVs of FDG uptake in PET may be a noninvasive biomarker for advanced CRC, indicative of a large hypoxic tumor with deep invasion. |
| head and neck cancer | OBJECTIVE: The aim of this study was to determine the biological significance of glucose transporter (Glut)-1 and Glut-3 expression in head and neck carcinoma (HNC). METHODS: We detected expression of Glut-1 and -3 in 38 HNCs and analyzed the relationship between increased expression and the biological behavior of HNCs. RESULTS: The gene expression levels of Glut-1 and -3 in HNCs were significantly higher than those in adjacent cancer tissues or in normal tissues. The GLUT-1 gene level was correlated with the lymph node metastasis and clinical stage of 38 HNCs, and the GLUT-3 gene level was correlated with the lymph node metastasis of 38 HNCs. Of the 38 cases, 30 showed positive expression of Glut-1 protein. The Glut-1 protein expression level was related to the lymph node metastasis and clinical stage of 38 HNCs. However, there was no case that expressed Glut-3 protein. The high expression of Glut-1 gene and protein was associated with poor survival in the HNCs. GLUT-3 gene expression was not associated with the prognosis of HNCs. CONCLUSIONS: GLUT-1 gene expression level and protein expression were correlated with lymph node metastasis, poor survival and clinical stage of HNCs. The GLUT-3 gene expression level was high in HNCs, and its expression was associated with an increased incidence of lymph node metastasis of HNCs. |
| laryngeal cancer | OBJECTIVE: To investigate the expression of glucose transporter 1 (GLUT1) in human laryngeal squamous cell carcinoma and its relationship to clinical pathologic factors. METHOD: tumor tissues and the peritumoral and normal laryngeal tissue were collected from 57 patients with laryngeal squamous cell carcinoma. The expression of GLUT1 was evaluated at protein and mRNA level by immunohistochemistry and reverse transcription polymerase chain reaction(RT-PCR) in relation to clinical pathologic characteristics. RESULT: GLUT1 protein and mRNA expression were significantly higher in tumoral tissues than in peritumoral and normal tissues (P < 0.01, respectively). Poorly differentiated tumor expressed higher levels of GLUT1 protein and mRNA compared to well differentiated ones (P < 0.05, respectively). Stronger GLUT1 protein and mRNA were detected in larger tumors compared with that in smaller ones (P < 0.05, respectively). There were significant differences in GLUT1 protein and mRNA expression in relation to TNM stage (P < 0.05, respectively). The expression levels of GLUT1 protein and mRNA were positively correlated with lymphatic metastasis (P < 0.05, respectively). CONCLUSION: GLUT1 may play an important role in tumorigenesis, development, differentiation, lymphatic metastasis and prognosis of human laryngeal squamous cell carcinoma, and may be a useful marker of diagnosis and prognosis. |
| laryngeal cancer | OBJECTIVE: To detect the expression of hypoxia inducible factor 1 alpha (HIF-1alpha), glucose transporter protein-1 (GLUT-1) and vascular endothelial growth factor (VEGF) in human laryngeal carcinoma tissue, and to study the relationship between hypoxia and HIF-1alpha, GLUT-1, VEGF in human laryngeal carcinoma Hep-2 cells and to explore the effect of HIF-1alpha, GLUT-1 and VEGF as endogenous hypoxic markers on laryngeal carcinoma. METHODS: The expression levels of HIF-1alpha, GLUT-1 and VEGF were detected in 35 cases of laryngeal carcinoma by SP immunohistochemical methods and in Hep-2 cells by SP immunocytochemical methods. The relationship between HIF-1alpha and GLUT-1, VEGF protein expression was analyzed. RESULTS: Of the 35 cases, 16 cases expressed HIF-1alpha, 16 cases expressed GLUT-1, 19 cases expressed VEGF. The expression of HIF-1alpha and VEGF were closely correlated with pathologic grading and lymphnode metastasis. GLUT-1 was correlated with lymphnode metastasis. The expression levels of HIF-1alpha, GLUT-1 and VEGF in Hep-2 cells under hypoxic condition were higher than those under normoxic condition. CONCLUSION: HIF-1alpha may promote the expression of GLUT-1 and VEGF in laryngeal carcinoma, furthermore promote tumor angiogenesis, invasion, and metastasis of the laryngeal carcinoma. |
| oral cancer | OBJECT: Malignant cells show increased uptake, which is considered to be facilitated by glucose transporters (GLUTs). Increased GLUT-1 expression has been reported in many human cancers. We hypothesized that a oral squamous cell carcinoma, characterized by high frequency of lymph node metastasis, distant metastasis or local recurrences, was associated with GLUT-1 overexpression in invasion front. METHODS: GLUT-1 immunostaining in invasion front was studied on 24 oral squamous cell carcinomas, and revealed the correlation with the clinical characteristics. RESULT: The analysis showed that all oral squamous cell carcinoma patients and GLUT-1 expression correlated the depth of the tumors (P = 0.023 < 0.05). Furthermore the survival of patients who had overexpression of invasion front was significant shorter than that of patients with GLUT-1 weakly positive (P = 0.046 < 0.05). No significant association was noted between GLUT-1 immunostaining and either age, gender, subsites, tumor size, or lymph node status. CONCLUSION: The present study shows that GLUT-1 served as a marker indicating that tumors with deep invasion tended to result in a worse prognosis in patients due to either lymph node metastasis, a recurrence of the primary lesion or distant metastasis. |
| papillary cancer | Glucose transporter-1 (GLUT-1) expression was immunohistochemically analysed in a total of 268 cases of thyroid gland disease, including 129 cases of papillary carcinoma (PC), 60 cases of follicular carcinoma (FC), 57 cases of follicular adenoma, and 22 cases of adenomatous goitre. Seventy-one percent (91/129) of PC cases showed GLUT-1 expression, semi-quantitatively evaluated as: +, 21 cases (16%); 2+, 37 cases (29%); 3+, 33 cases (26%); and negative, 38 cases (29%). These positive cases were divided into two groups: 'membrane-like' pattern in 24 cases (19%), and 'cytoplasm-predominance' pattern in 67 cases (52%). GLUT-1 expression was observed in 5% (3/60) of FC cases, but all follicular adenomas and adenomatous goitres were negative for GLUT-1 (PC vs. FC, p<0.0001). Membrane-like expression was observed more frequently in non-organ-confined PCs (pT4) than in organ-confined PCs (pT1, 2, and 3) (p=0.0056). Seventy-five percent (18/24) of PC cases showing membrane-like expression were non-organ-confined. The membrane-like pattern was observed more frequently in PCs with lymph node (LN) metastasis compared to those without (p=0.0036). Ninety-two percent (22/24) of PC cases showing the membrane-like pattern were not organ-confined. Semi-quantitative analysis of glut-1 mRNA by RT-PCR showed a tendency toward higher expression in PCs compared to FCs, follicular adenomas and adenomatous goitres, and the mRNA expression in PCs with a membrane-like pattern was higher than those showing cytoplasm-predominance. We concluded that: 1) GLUT-1 is immunohistochemically useful in distinguishing PC from FC and benign diseases; 2) GLUT-1 may play an important role in the advancement of PC and LN metastasis, and its membrane-like expression is of more clinical significance than the cytoplasm-predominance pattern; and 3) glut-1 mRNA expression corresponds with the immunohistochemical expression profile. |
